Introduction
Nowadays, genosomes is also known as cationic lipid-DNA complexes or lipoplexes, becomes one of the most promising nonviral DNA-delivery systems, particularly in gene transfer therapy. Different from the virus-based methods, cationic liposomes offer biocompatibility, biodegradability, and nonimmunogenity in carrying DNA. The mechanism of interaction of DNA and the cationic surfactant attributes to the formation of lipoplexes is mostly electrostatic, between the negatively charged DNA and the positively charged cationic liposomes. In addition, it is an entropically driven process that comes from the generation of counterions to the solution. Neutralization of the negative charges of DNA and cationic liposomes leads to not only the complexation of DNA but also the formation of different more compact and condensed structures. This compaction is presented on the surface of the cationic liposome. Additionally, the optimum morphology of the genosomes is mainly depending on the elasticity and the surface charge density, which are associated with the nature, concentration, and composition of the cationic-zwitterionic lipid mixture, pH, temperature, and ionic strength.
